Antibacterial effect of phenothiazines against Klebsiella pneumoniae

Klebsiella pneumoniae is a gram-negative, encapsulated, non-motile bacterium found in the environment and has been associated with infection in immunocompromised patients. K. pneumoniae strains have become increasingly resistant to antibiotics, rendering infection by these strains very challenging to treat. Notably, multidrug resistance Klebsiella pneumoniae infections are responsible for the increasing number of mortality among hospitalized and immunocompromised patients.Phenothiazines (PTZ) are neuroleptics used to treat psychosis including schizophrenia; violent, agitated, disturbed behavior. We aimed to evaluat the antimicrobial acticvity and the synergism of PTZ with antimicrobial peptides against MDR K. pneumoniae. Our results indicated that PTZ, specifically chlorpromazine (CPZ) effectively inhibited the growth and the biofilm against K. pneumoniae. The reduced efflux activity may contribute in part to the antibacterial activity of CPZ. In addition CPZ synergized the antibactericidal activity of polymyxin B and colistin against tested bacteria strains.
